Frankly speaking, Robert Holton is kinda of "Taxol Special" synthetic chemist
for a long time. It's unfair to compare him & KC only by this molecule.
In the last 10, maybe 15 years, Taxol has occupied most of Holton group's
attention and effort...
An issue of C&EN News in 2003 higlighted Holton's contribution
to taxol semisynthesis which will provide the commercial supply...
Refer:
http://www.chem.fsu.edu/faculty/rah.htm http://pubs.acs.org/cen/coverstory/8137/8137taxol.htmlThe low yield of KC's synthesis of Taxol is mainly due to the unpredictable
low yield (approximatly 20%) of McMurry reaction which is used to close
Ring-B... That impressed the overall yield a lot...
But after a plan of total synthesis was designed, it's very hard to replace
the reaction of a crucial step with another one....

: Nicolaou合成taxol 39步 0.09%的产率
: Holton59步,0.3%
